Microfluorometric assay for quantification of anti-erythrocytic antibody level in sera of Plasmodium berghei ANKA infected semi-immune mice

چکیده
A simple, sensitive and reproducible fluorometry method was developed to quantify anti-erythrocytic auto-antibody in the sera of semi-immune mice. The level of anti-erythrocytic auto-antibody measured using fluorometry was significantly higher in sera of Plasmodium berghei ANKA infected mice than the uninfected, p< 0.0001. This result correlates significantly with the standard ELISA assay, p<0.0001, r=0.7766. Both methods show significant correlation with regard to extent of Hb loss, p<0.006 (r=0.33) (fluorometry method) and p< 0.0001, r=0.30 (ELISA). Thus the fluorometry method may serve as an alternative to standard ELISA assay and can be modified to suit similar objectives.

Acute Toxicity Studies and Anti-plasmodial Potentials of Newbouldia laevis and Crateva adansonii in Plasmodium Berghei-infected Mice
Background: Newbouldia laevis and Crateva adansonii are popular plants used traditionally in the management of patients with malaria infection in Nigeria. In this study, the acute toxicity and anti-plasmodial effects of the leaf extracts of Newbouldia laevis and Crateva adansonii were investigated in mice. Methods: The Anti-plasmodial activities of both extracts were investigated individual...
متن کاملInfection with Plasmodium berghei boosts antibody responses primed by a DNA vaccine encoding gametocyte antigen Pbs48/45.
An important consideration in the development of a malaria vaccine for individuals living in areas of endemicity is whether vaccine-elicited immune responses can be boosted by natural infection. To investigate this question, we used Plasmodium berghei ANKA blood-stage parasites for the infection of mice that were previously immunized with a DNA vaccine encoding the P. berghei sexual-stage antig...
متن کاملComparative Anti-malarial and Toxicological Properties of the Stem Bark Extracts of Nauclea latifolia and Terminalia glaucescens Against Plasmodium berghei-infected Mice
Background: Anti-plasmodial activities of the methanol stem bark extracts of Nauclea latifolia, and Terminalia glaucescens were investigated in mice infected with Plasmodium berghei. Methods: A total of 24 Plasmodium berghei infected (P. berghei infected) mice were divided into 8 groups of 3 each. Groups A-F were given 100, 300 or 500 mg/kg of either extracts. Groups G and H received 2 mL norm...

Objective: Vernonia amygdalina (V. amygdalina) leaf is locally employed in the Southern region of Nigeria in the treatment of malari a infection. This study evaluated the in vivo antiplasmodial, antioxidant and immunomodulatory effect of ethanol extract of V. amygdalina leaf. Materials and Methods: The active principles of the dried leaf were extracted with ethanol. For quality validation, ch...
متن کاملEstradiol, but not dehydroepiandrosterone, decreases parasitemia and increases the incidence of cerebral malaria and the mortality in plasmodium berghei ANKA-infected CBA mice.
OBJECTIVE The effect of castration and subsequent replacement of dehydroepiandrosterone (DHEA) or estradiol on parasitemia, mortality and incidence of cerebral malaria (CM) was evaluated in CBA mice infected with Plasmodium berghei ANKA. METHODS Female mice were castrated, and groups of 12-15 animals received daily injections of DHEA, estradiol or saline. Four days after the start of treatmen...
